真诚请教免疫组化复染的时间掌握。我用苏木精10秒，盐酸酒精5秒，碳酸理5秒，每一步后都用了水洗，但总掌握不好。作细胞凋亡也是，说明书说用苏木精淡染，但我染后虽然其他胞核蓝染极度轻微，但凋亡的染色处就没有没有复染前漂亮。
大家能否把自己的复染经验介绍一下？

我也有这个问题，我一般用苏木精3分钟，盐酸分化液10秒，然后就脱水等。为什么coldant 你要用碳酸锂？？？
还有一个问题，是否一定要做复染？究竟如何决定是否该复染？希望获得解答？？

复染主要是让胞核蓝染，以衬托胞浆或者胞膜的DAB阳性。为什么用碳酸理我也不知道，只是大家都这样做的。我觉得你的苏木精3min太长了，

可我认为复染有时会掩盖掉一些阳性染色，有种两重性的作用，既可衬染，又有掩盖作用。
碳酸锂不会是“大家都这样做的”的吧？？？我们这儿没听说呀？？？我真的很奇怪？？？
至于苏木精，我其实有时用到4min，可能你的苏木精效果特别好吧。我用4min也不是很蓝呢。

Hi everybody,
I think most of Lab. using Harris’ Hematoxylin, it is very strong staining nuclear of cell.  So for IHC counter staining, I just stain 3 Second and washing to blue.  You also can choose Gill’s and Mayer’s Hematoxylin.  I tried using 0.1% Methyl Green or Fast red; they are very good for counterstaining.

I used different hematoxylin.  I know most Pathology and Histology Lab. using Harris' Hematoxylin for HE staining.  You can use it, at 1:10 dilution and for 3 second or more.  Please try different time and find optimal staining time.
